Achyranthes bidentata
This product is the dried root of Achyranthes bidentata Bl. of the Achyranthes family. It is dug up in winter when the stems and leaves wither, the fibrous roots and mud are removed, and the roots are bundled into small bundles. After the bundles are dried and wrinkled, the tops are cut evenly and dried.
This product is slender and cylindrical, straight or slightly curved, 15 to 70 cm long, 0.4 to 1 cm in diameter. The surface is grayish yellow or light brown, with slightly twisted fine longitudinal wrinkles, sparsely arranged lateral root marks and transverse long lenticel-like protrusions. It is hard and brittle, easy to break, and becomes soft after being damp. The cross section is flat, light brown, slightly horny and oily. The central vascular bundle has a large xylem, yellow-white, and its periphery is scattered with many yellow-white dot-like vascular bundles, which are arranged in 2 to 4 rounds intermittently. It has a faint smell and tastes slightly sweet and slightly bitter.
(1) Cross-section of this product: The cork layer is composed of several rows of flat cells, extending tangentially. The inner cork layer is narrower. The heteromorphic vascular bundles are tough outside and arranged in 2 to 4 whorls. The vascular bundles in the outermost whorl are smaller, and some have only 1 to several vessels. The inter-bundle cambium is connected into a ring, and the inward vascular bundles are larger; the xylem is mainly composed of vessels and small wood fibers, and the xylem in the center of the root is integrated into 2-3 groups. The parenchyma cells contain calcium oxalate sand crystals.
(2) Take 4g of the powder of this product, add 50ml of 80% methanol, heat and reflux for 3 hours, filter, evaporate the filtrate to dryness, add 15ml of water to the residue, dissolve it with slight heat, add it to a D101 macroporous adsorption resin column (inner diameter 1.5cm, column height 15cm), elute with 100ml of water, discard the water, elute with 100ml of 20% ethanol, discard the eluate, elute with 100ml of 80% ethanol, collect the eluate, evaporate to dryness, add 1 1ml of 80% methanol to dissolve the residue, and use it as the test solution. Take 4g of Achyranthes bidentata reference medicinal material and prepare reference medicinal material solution in the same way. Take 0-ecdysone reference material and ginsenoside R. Reference material and add methanol to prepare solutions containing 1mg per 1ml as reference material solutions. According to the thin layer chromatography method (General Rule 0502), take 4~8μl of the test solution, 4μl of the reference medicinal material solution and the reference material solution, and spot them on the same silica gel G thin layer plate, using chloroform-methanol-water-formic acid (7:3:0.5:0.05) as the developing agent, develop, take out, dry, spray with 5% vanillin sulfuric acid solution, and heat at 105℃ until the spots are clearly colored. In the chromatogram of the test sample, spots of the same color appear at the corresponding positions of the chromatogram of the reference medicinal material and the chromatogram of the reference material.
Moisture content must not exceed 15.0% (General Rule 0832 Second Method).
The total ash content shall not exceed 9.0% (General Rule 2302).
The residual amount of sulfur dioxide shall be measured according to the determination method of residual sulfur dioxide (General Chapter 2331) and shall not exceed 400mg/kg.
According to the hot soak method under the determination method of alcohol-soluble leachables (General Chapter 2201), water-saturated n-butanol is used as the solvent, and it should not be less than 6.5%.
Determine according to high performance liquid chromatography (General Chapter 0512).

Determination method: Precisely draw 10μ each of the reference solution and the test solution, inject them into the liquid chromatograph, and measure. Calculated as a dry product, this product contains no less than 0.030% of 0-ecdysone (C27 H44O7).
